Photo-oxidation and Quinacrine Fluorescence Response in the Mitotic Chromosomes of the Flesh-flies Sarcophaga
نویسنده
چکیده
Experiments have been carried out to investigate the DNA base specificity of lightinduced banding (LIB) produced by photo-oxidation of chromosomes followed by Quinacrine staining to detect denatured DNA. In the present study mitotic preparations from Sarcophaga ruficornis and Sarcophaga argyrostoma were used to detect the visible light and fluorochrome interactions. Our results indicated that specific destruction of guanine residues is the main effect of photo-oxidation and LIB is a base-specific phenomenon. In addition, photo-oxidation may also cause DNA-protein cross-linking which affects the binding of some dyes, while prolonged photo-oxidation appears to cause more general damage to DNA.

Myiasis is always resulted by depositionof egg or young larvae of cyclorrhapha flies’ in live tissus of human or animals, which follows by their Development. Although the survival of mentioned larves is depended on existance of livestock and wild animals, the flies can be seen in parks, dairy farms and certain plants and also solid wasts waiting for proper hosts. Background and purpose: One of the most popular methods to control the synanthropic flies is using parasitoid wasps. The aim of this study was to estimate the experimental parasitism rates of pupae of Musca domestica, Lucilia sericata, and Sarcophaga heamorrhoidalis by parasitoid wasps, including Nasonia vitripennis, Spalangia nigroaenea, and Pachycrepoideus vindemmiae. متن کاملCharacterization and origin of extrachromosomal DNA granules in Sarcophaga bullata.
We have used endonuclease treatment in situ, followed by Giemsa or ethidium bromide staining, for mapping repetitive sequences on the chromosomes of the flesh fly Sarcophaga bullata and thus for studying extrachromosomal DNA granules in this species. All three restriction enzymes employed (HaeIII, A1uI and HindIII) show the same cytological effects, except for a single interstitial band. In bot...
